Concluding remarks and future perspectives
Such considerations as a whole support the possibility that COVID-19 mRNA vaccines under some circumstances induce high and possibly toxic amounts of S protein in organs and tissues, in turn leaking into the circulation. In animal models, it is well established that lipid nanoparticle-carried mRNAs undergo systemic disposition and expression in organs such as liver, skeletal muscle, and lungs [13]. It can be suggested that at least part of the risk to develop adverse reactions following vaccination with mRNA products depends on the organs/tissues where S protein production occurs, as well as on the total amount produced and on the production time course. For example, it was established long ago that distinct tissues widely differ in the efficiency of protein synthesis, but no one so far assessed whether and to what extent this is relevant for the efficacy and safety of mRNA vaccines [13]. Therefore, we further recommend careful characterization of COVID-19 vaccines with regards to systemic disposition, including organs and tissues where S protein production occurs and interindividual variability in local protein synthesis efficiency, to provide a rational basis for dose individualization and to identify subjects at risk for adverse reactions due to either vaccine-induced S protein production in vulnerable sites, excessive S protein production, or both


Overall my conclusion is that now the emergency situation regarding the covid epidemic has subsided, much more research is needed before continued widespread use of mRNA vaccines in large non vulnerable populations.

Here is a paper from August 2022 looking at the pharmacology of mRNA vaccines suggesting that the way they work is more akin to pharmaceutical drugs than conventional vaccines with significant implications for post marketing safety surveillance.

Skimming the linked articles, they are careful to say "Covid-19 mRNA vaccines" and not simply "mRNA vaccines".

Since mRNA vaccines have been proposed, and are likely to be released, for other diseases this is an important distinction. The lipids, salts, sugars and even the individual nucleotides can be regarded as "inactive ingredients". Inactive ingredients can be harmful, and that has and should continue to be tested and safer replacements used if possible. But the inactive ingredients should be the same in vaccines for different diseases so the safety of Covid mRNA vaccines directly informs this aspect mRNA vaccines for other diseases.

The active ingredient of an mRNA vaccine is the sequence of nucleotides and indirectly the protein they encode. This has several implications.

First, all the first generation covid vaccines use the same spike protein. If this is a problem, say the S protein cause an allergic reaction or interferes with cellular chemistry, then this needs to be checked for all the covid vaccines, and indeed for catching covid, since an infection will release S-protein. There may be differences between different covid vaccines due to dosage, or site of delivery but it may be possible to alter these for all covid vaccines, including mRNA ones.

Second, the active ingredient of mRNA vaccines for different diseases are completely different, and you need to run complete tests for every different vaccine. The safety of covid mRNA vaccine says little about the safety of a flu mRNA vaccine. In theory mRNA could code for a protein that is a powerful neurotoxin or allergen. Someday some wretched person will probably do that on purpose, but it would be worse to do it by accident.

Third, it would be good to change the title of this thread.
